His college success propelled him into the NFL, where the Indianapolis Colts selected him in the third round of the 2012 draft, making him the fourth football player from FIU to be drafted into the league.

As a Colt, Hilton carved a niche for himself as one of the top wide receivers in the league. His NFL career would see him selected to four consecutive Pro Bowls from 2014 to 2017. He even led the league in receiving yards in 2016, finishing with an astounding 1,448 yards. Over ten seasons with the Colts, he accumulated a total of 638 receptions, 9,812 receiving yards, and 53 touchdowns.
